国内菜用黄花菜种质资源遗传多样性分析

Genetic Diversity of Vegetable Daylily Germplasms in China

摘要: 对28份国内栽培的黄花菜资源19个形态学特征进行了遗传多样性研究,并且利用SCoT分子标记技术对这些黄花菜资源和3份新资源进行遗传多样性分析。结果表明:农艺性状分析显示种质间数量指标的变异程度较大,10个数量指标的变异系数为14.5%~37.2%,其中最大的为分蘖数(37.2%),其次为叶宽(35.5%),说明这2个性状的变异程度较大,性状分离明显;叶长(14.5%)最小,说明叶长在种质间较为稳定,变异程度小;形态学和分子标记聚类结果相似,均把黄花菜资源分成2类,而且每类的资源是一致的,3份新资源归类到I类(红花);另外,形态学不能区分的五月花和六月花两份资源,利用SCoT标记技术可以很好地区分。研究表明,形态学和SCoT分子标记均可作为黄花菜遗传多样性分析的工具,但是SCoT标记技术更加快速、更加灵敏、更加准确。

Abstract: The genetic diversity of 28 vegetable daylily germplasms in China were studied based on 19 morphological characteristics as well as the start codon targeted polymorphism(SCoT) markers of the plants. The agronomic analysis showed that the variations on 10 quantitative indices among the germplasms were large with the variation coefficients ranging from 14.5% to 37.2%. The highest coefficient was found on tiller number, followed by leaf width at 35.5%, while leaf length being the lowest. The results obtained from the molecular marker clustering were similar, and both methods classified the germplasms into two classes.However, the SCoT technique clustered the 3 newly introduced germplasms into Class I (red flower) and could satisfactorily distinguish Wu Yue Hua from Liu Yue Hua while the morphological approach failed to do so. It appeared that the utilization of SCoT markers was faster, more sensitive and accurate than that of the agronomic properties for the identification of the daylily cultivars among various germplasms.
